Due to the Gold Rush.
The California Gold Rush took place in 1848-1849 and was the primary reason California was settled by Americans.
San Francisco became the major outlet to the rest of the world. It was too timely to travel across land back to the east, so all commerce generated from the Gold Rush went through San Francisco, the biggest port city on the west coast. The term 49er essentially refers to the miners, explorers, and fortune hunters who came to California during this time.
